Doctorant (H/F) pour l’identification et la caractérisation de complexes ribonucléoprotéiques impliqués dans la réplication des virus de la Dengue et du Zika

Le doctorant travaillera sur une étude qui permettra de mieux comprendre les mécanismes de la réplication virale afin de contribuer à l'élaboration rationnelle de nouveaux antiviraux. Les objectifs de ce projet de thèse sont de: (i) identifier et caractériser les facteurs chez le moustique qui interagissent avec l'ARN génomique de ces virus et (ii) caractériser au niveau structural et biophysique plusieurs complexes ribonucléoprotéiques impliquant des régions non codantes du génome viral et des protéines de l'hôte moustique et nécessaires au le cycle de réplication viral. Dans ce but, des approches biophysiques intégrées (microcalorimétrie, biosenseur à puce ADN, spectrométrie UV-visible, Spectrométrie de Masse, Cryo-Microscopie Electronique, Résonance Magnétique Nucléaire et Cristallographie aux rayons X) seront utilisées. Ce projet sera également réalisé en collaboration avec l'Université du Minas Gerais au Brésil, spécialisée de longue date dans les infections des virus de la Dengue et du Zika, et fera appel à l'Insectarium de l'IBMC afin d'identifier d'éventuelles protéines de l'hôte importantes pour le cycle viral.

We are looking for a PhD student in Molecular Biology to participate in a research programme. The thesis project will focus on the study of the translation of the messenger RNA encoding the Tau protein. The study will include the use of cell-free translation extracts prepared from several cell types relevant to Alzheimer's disease. In particular, the project will focus on the synthesis of N-terminally shorter variants of the Tau protein.

D. student (M/F) for the development of depollution enzymes by directed evolution and functional screening in microfluidic systems. The thesis work will be carried out within the "Digital RNA Biology" team (Institute of Molecular and Cellular Biology, Strasbourg). To do this, two complementary strategies will be used in parallel, in close collaboration with the team of Prof. Stéphane Vuilleumier (University of Strasbourg). Firstly, bacteria from samples taken from sites contaminated by fluorinated compounds will be individualised in water-in-oil droplets of a few picolitres containing a minimum synthetic culture medium supplemented with the fluorinated compound to be degraded and used as a carbon source. Thus, the ability of the organism in the droplet to degrade the target compound will result not only in its growth, but also in the release of fluoride. The latter will be detected by an RNA molecular probe recently developed by the team, which converts the presence of fluoride ions into fluorescence emission.
