Deciphering complex molecular mechanisms

Deciphering molecular mechanisms at work in the cell requires the preparation and the testing of several mutants, a time consuming and labor-intensive work. Nevertheless, our ultrahigh-throughput screening pipeline allows to generate and functionally screen millions of mutants of a target RNA in a single day. The analysis of the sequence content of the initial and the selected libraries with Next Generation Sequencing drastically speeds up the mutational analysis. We recently applied this approach to decipher eukaryotic translation initiation.
