The mission of the project will consist in the analysis of RNA modifications (mainly tRNA and rRNA) in different strains of Staphylococcus aureus and in different stress conditions encountered during infections. This mapping will be performed using innovative methodologies, including mass spectrometry and high-throughput sequencing. The impact of variations in the epitranscriptome on the translation process and on the decoding rules will be analyzed at global scale by means of “ribosome profiling” and in details by in vitro translation systems. Based on the obtained results, mutations in key modifying enzymes will be analyzed for their function in the physiology and pathogenicity of S. aureus.
